In peptide bond the amino group of one amino acid reacts with carboxylic acid group of other amino acid.

Now in His-Gly-Ala, Histidine has a free amino group and its carboxylic group forms bond with amino group of Glycine. The carboxylic group of glycine forms bond with amino group of alanine. The carboxylic group of alanine is free in this tripeptide.

While in Ala—Gly—His ,alanine has a free amino group and its carboxylic group forms bond with amino group of Glycine. The carboxylic group of glycine forms bond with amino group of histidien. The carboxylic group of histidine is free in this tripeptide.
